Sidec Technologies AB, Sweden, today announced a contract research
collaboration with Procognia Ltd, UK, to locate protein glycosylation
sites.
Sidec’s 3-D reconstruction technology for protein imaging will be used
to determine the location of glycosylation sites on proteins supplied
by Procognia.
Sidec has developed a technology for 3-D reconstruction of multiple
protein structures in a single analysis. This technology has the
potential to provide a rapid tool for locating glycans on intact
glycosylated proteins. Current glycan localization processes requires
cleavage of the protein and extensive purification and analysis of the
protein fragments.
It is well known that differences in the glycosylation of proteins can
have profound effects on their biological properties. Yet, the
glycosylation on biopharmaceutical products is one of the most
difficult manufacturing processes to control. Procognia has developed
a technology for the rapid analysis of glycosylation patterns of
intact proteins in a mixture of glycoforms.
If the 3-D technology is successful in visualizing the location of
glycans on intact proteins, it may obviate more complicated analyses
required to obtain that information using current technology.
Therefore, Procognia’s technology could be complimented by a means of
rapidly determining the location of the glycosylation sites. Sidec’s
3-D reconstruction with SET (Sidec™ Electron Tomography) coupled with
Procognia’s U-c fingerprint of protein glycosylation patterns has the
potential to be important in the discovery, development and
manufacture of protein-based pharmaceuticals.
